PREPARATION OF TISSUES FOR SEM

Dehydration:

Fix tissue immediately after removal from organism in 2% glutaraldehyde in Millonig's Buffer (MB) for at least overnight. The sample can remain indefinitely in this fixative.
Rinse tissue in fresh MB 3 x 15 min.
Sequentially dehydrate speciments with acetone for 10 min in each of the following following:

	% acetone (v/v in Type I water)		
		25
		50	
		70
		90
		95
		100
		100
		100 (optional)

Go directly from the last acetone exhange to critical point drying.
